First, you’ll need to locate the giant Joker Face in the main hub. This is easy to find, but you won't have access to it from the start of the game. The Joker Face can be found once you've completed the game's first chapter (the one that ends with The Joker escaping in a helicopter). You need to walk up to it and press circle/B. What you then do is spell out the codes on his teeth. Each code is 5 letters long. Once you have entered the code, the character will be unlocked.
